Systematics Section / ASPT Prince, Linda [1]. Estimating species level phylogenies in Bromeliaceae: an example from Guzmania. ESTIMATION of species relationships within Bromeliaceae using phylogenetic analysis of nucleic acid sequence data has not utilized rapidly evolving regions of the nuclear and plastid genomes. To date, there are no sequences of the nuclear internal transcribed spacer regions (nrITS1 and nrITS2) in GenBank, presumably due to difficulty amplifying and sequencing this GC-rich region (up to 78%) in Bromeliads. Plastid non-coding regions such as the atpB-rbcL and trnL-trnF intergenic spacer regions (IGS), and the rps16 and trnL introns have been used for intrageneric studies. These plastid regions are not evolving very quickly, thus provide little resolution between closely related taxa. This study focused on improving methods for obtaining nrITS data, and compared those data to sequences from over 20 chloroplast IGS regions and introns, assessing the phylogenetic utility of each for their effectiveness at elucidating relationships among a group of closely related species of Guzmania. Analysis of the molecular data confirms the monophyly of the Guzmania (23 ingroup taxa) and result in many resolved branches with moderate bootstrap and posterior probability support.
